Here applicant should show confidence and de-emphasize the past and show what he/she can offer for the company in future. Employers are not always right, if you put forward the good reasons and explain the situation briefly and say that you had a difference of opinion with your previous employer. Then, this would be consider as a positive attitude for showing honesty, but be sure to be brief while explaining the situation and do not drag it at any cost and move on for the next question fast.

Are you open to work on nights and weekends, if so…what interests you to do that?

Here, in this case it actually depends on each candidate. If he/she is fine with working on night shifts and on weekends, he/she can simply answer that he/she is a work alcoholic and their family clearly understands their priority at the moment and would co-operate in this timings. So here, you are almost close to getting the job as interview would be happy to know that they will have people to work on night shifts or during weekends. Incase if applicant is more of a balanced life person, then he/she needs to answer this question tactfully by asking if interviewer has any top performers working in a reasonable time, or if they have any norm for the best top performers and can add in that applicant would get into that category from his/her good performance. He/She could also add in that he/she would like to go and see his/her family after the work at the end of the day. Say that, seeing my family during weekends adds richness in the quality of my work, and ask if there are any chances to handle some amount of work to carry it home. He/She would love to do that and  ensure balancing life will not effect a bit on their professional work.
